Predicting the Environmental Hazards of Xenobiotics by QSPR Methods I. Ethylene Oxide - The paper presents the combined application of QSAR expert systems to predict the environmental hazard of ethylene oxide and its metabolites. The metabolic tree was generated in four metabolic levels by the MetabolExpert Ver. 10.0 program. The toxicity of ethylene oxide and its 24 metabolites was then subjected to toxicology prediction study using the HazardExpert program. Ethylene oxide is found highly toxic in four of the seven toxicity classes, such as oncogenic, mutagenic, membrane irritating and teratogenic. The computed results are validated by experimental data. Program database was updated by recent results on sensitisation and neurotoxicity. The toxicity of ethylene glycol, the first and largest metabolite, compared to the ethylene oxide is low (LD50 oral rat 4700 vs. 72 mg kg-1), and metabolises rapidly in water. Most of the other metabolites are shown essentially harmless for the environment. Teratogenic effects are predicted strongly for ethylene oxide but epidemiological studies have not been reported on the subject, yet. The potential hazards of industrial activities should be assessed in the very early stages of technology design, much before commercialisation whether a new technology or just a technology modification is to be invested. Assuming properly updated database, the use of MetabolExpert and HazardExpert provides a reliable, rapid prediction for environmental hazards of chemicals and their metabolites of good informative value.
