Entwicklung zeitvarianter Verfahren der Bispektral- und Bikohärenzanalyse zur Detektion und Qualifizierung transienter quadratischer Phasenkopplungen

Diese Dissertation befasst sich mit der Entwicklung von Verfahren der zeitvarianten Bispektral- und Bikohärenzanalyse zur Detektion und Quantifizierung transienter quadratischer Phasenkopplungen (QPC) auf der Basis der indirekten Schätzung (Fourier-Transformation der Momente 3. Ordnung). Ausgangspunkt ist die adaptiv rekursive Momentenschätzung 3. Ordnung, deren Schätzeigenschaften verbessert werden. Als Ergebnis werden geglättete Pseudo-Zeit-Frequenz-Verteilungen 3.Ordnung (3rd order SPTFD) eingeführt, die u.a. die untersuchten Schätzer (Rihaczek-, Wigner- und modifiziertes Wigner-Spektrum) integrieren und mit Hilfe der lokalen Momentenfunktion, des Zeitglättungs- und des Lag-Fensters parametrisieren. Solche Methoden gewährleisten eine kontinuierliche Analyse fortlaufender Signale bei voneinander unabhängig einstellbarer Zeit- und Frequenzauflösung. Bekannte Bikohärenzschätzer sind zur Normierung indirekt geschätzter Bispektren nicht geeignet. Deshalb wird als Grundlage der zeitvarianten Bikohärenzschätzung eine neue, modifizierte Normierungsform eingeführt. Die Eigenschften der entwickelten Methoden werden umfassend analysiert. Neben den statistischen Eigenschaften (Signifikanzgrenzen, Bias, Varianz) und den Auflösungeigenschaften werden unter anderem der Einfluss des Signal-Rausch-Abstandes und die Prinzipien der Termlokalisation und Termunterdrückung untersucht. Die Methodik wird zur Analyse transienter quadratischer Phasenkopplungen der Burst-Aktivitäten im trace alternant EEG gesunder Neugeborener und im Burst-Suppressions-EEG sedierter Intensivtherapie-Patienten eingesetzt. Die Ergebnisse bestätigen, dass Burst-Aktivität im diskontinuierlichen EEG das Produkt nichtlinearer Generatorprozesse ist. Quadratische Phasenkopplungen sind nachweisbar, deren Stärke im zeitlichen Zusammenhang mit Erscheinen und Verlauf der Burst-Aktivität steht. Mit Hilfe der zeitvarianten Wigner-Phasenbikohärenz werden im Blutvolumenpulssignal von Patienten mit peripherer arterieller Verschlusskrankheit (paVK) und Diabetes-Patienten charakteristische Merkmale gefunden, in denen sich diese Patienten von Gesunden unterscheiden. Diese werden zu einem singulären Bispektralindex zusammengefasst. Dessen diagnostische Relevanz wird an 27 Probanden und 33 Patienten untersucht.
This thesis deals with the development of methods of time variant bispectral and bicoherence analysis for the detection and quantification of transient quadratic phase coupling (QPC) based on an indirect estimation approach (Fourier transform of the 3rd-order moments). The methodical origin is the adaptive recursive estimation of the 3rd-order moments. This approach is continuously advanced in order to improve the estimation properties. As a result, the class of 3rd-order smoothed pseudo time frequency distributions (3rd-order SPTFD) is introduced which includes and parameterises the investigated estimators (Rihaczek bispectrum, Wigner bispectrum and modified Wigner bispectrum) based on the local moment function, time smoothing window and lag window. Methods of the 3rd-order SPTFD allow an appropriate bispectral analysis of ongoing signals including independent adjustment of time and frequency resolution. Common bicoherence estimators are inappropriate for the normalisation of indirectly estimated bispectra. Therefore, a new modified normalising approach is introduced which represents the basis of the time variant bicoherence estimation. The estimation properties of the developed methods are analysed extensively. Statistical properties (significance thresholds, bias, variance), time and frequency resolution, the influence of the signal-to-noise ratio and the principles of term localisation and term suppression are investigated. The developed methods are applied to analyse transient quadratic phase couplings of burst activity in the trace alternant EEG of healthy newborns and in the burst suppression EEG of intensive care patients with neurosurgical deseases. The results confirm that burst activity in the discontinuous EEG is the result of nonlinear generation processes. Quadratic phase couplings can be verified and their degree corresponds with the appearance the time course of the burst activity. In another application, bispectral characteristcs of the pulse wave of diabetic patients and patients with peripheral arterial occlusive desease are detected by means of the time variant phase bicoherence. The characteristcs are combined into a singular bispectral index. Its diagnostic relevance is investigated based on 27 healthy subjects and 33 patients.
